  1. Services

Global metabolomics and integrative -omics (intOmics)

Metabolomics provides screening of targeted (~650) and untargeted (~2500) metabolites. Options for more detailed analyses include volcano plots, pathway enrichment and KEGG network optimization data. Metabolomic data can be integrated with (user provided) RNAseq using a metabolomics network analysis platform (IntOmix), for an optimized pathway analysis of relationships between metabolites and enzyme transcripts.

  2. Services

Metabolic flux and enrichment studies using MIMOSA

In this subcore, parameters of cellular and tissue metabolic flux are assessed by incorporation of stable isotope labeled tracers (e.g., 13C, 15N, 2D) into metabolism. The CMC uses the targeted mass isotopomer multi-ordinate spectral analysis (MIMOSA) platform to interpret stable isotope labeling patterns for calculation of rates of discrete steps in glycolytic and mitochondrial metabolism. Such measurements can be used to identify differences in fuel usage (e.g., glycolysis vs. beta-oxidation), and non-oxidative (reductive carboxylation, anaplerosis and cataplerosis) contributions to the TCA cycle.

  4. Instrument/Equipment

    3D printing

At the APC we have three 3D printers: a Formlabs Form 3, a Dremel 3D20, and a Lulzbot Taz Pro.

  5. Services

    Islet isolation and function

In this subcore, our technical expertise in isolating rodent islets including rats, mice and most recently ground squirrel islets, from the pancreas ex-vivo allows researchers to study the intact islets for their studies or to proceed with substrate stimulated insulin secretion functional studies using our BioRep Technologies Islet Perfusion Systems (8-channel and 12-channel) offered by our core. For non-rodent islet studies, human donor islet procurement assistance as well as unction studies are also offered.

  6. Services

Cellular and mitochondrial respiration and bioenergetics

Cellular and mitochondrial respiration are assessed with the Agilent Seahorse XF Pro. In a 96-well plate format, real-time measurements of cellular respiration, oxidative phosphorylation, mitochondrial leak, and mitochondrial transport are assessed as functional kinetic measurements of cellular bioenergetics. An integrated Agilent BioTek Cytation5 imager correlates cell number to respiration for more accurate normalization of respiration.

  8. Instrument/Equipment

    Abrasive water jet cutting

The high-pressure abrasive water jet cutter can be best thought of as a computer-controlled bandsaw. It is largely used to cut through flat material stock.
