About the instruments
The Core operates several sorters across four locations on YSM Campus: The Anlyan Center (TAC), Amistad, 300 George Street, and LEPH. The FACS Arias and Bigfoot are operated by trained staff and the Sony SH800 by users who have completed the necessary training.
Sort requests can be scheduled via PPMS. You must have a valid account and CoA on file to request a sort. Cancelations should be made via PPMS as well as a courtesy email to the sort operator if cancelation is within 48 hours of the requested sort time. Please refer to Policies to learn about our scheduling and cancelation rules.
Depending on the cell type being investigated, there are a variety of procedures for preparing cells. Before labeling cells for sorting, the antibody's concentration may need to be optimized, and appropriate negative and positive controls are needed. It is important to bring cells filtered and in single cell suspension, at proper concentration and stained with fluorochromes that are suitable for the cytometer of choice.
Cells have the flexibility to be sorted into either 5- or 15-ml tubes, as well as different plates, either as individual cells or multiple cells per well. They can also be placed onto microscopic slides for further examination. Moreover, it is worth noting that all sorters, except for the SONY SH800, have the capability to perform simultaneous 4-way sorting with the Bigfoot capable of 6-way sorting. Additionally, sorters are equipped to handle sorting live human, primate, or other potentially biohazardous cells.

Training & services
Do you need flow cytometry analysis, cell sorting, or both? Analysis results in data recorded by acquisition software. Cell sorting separates cells into pure populations and all the data acquired during sorting.
All scheduling, training requests, and services are made through an online portal, PPMS. To log onto the PPMS, point your browser to https://ppms.us/yale/start/. Your Yale NetID and password will serve as your account credentials. (Download a short guide to this intuitive login.) Please confirm your lab affiliation (“group”) and charging instructions. We also recommend that you check that your permissions for instrument access are correct. In most cases, details about your account may pre-populate, but new users and those without affiliated COAs will need to wait for the business office to approve the COAs you list before your account is functional. There might therefore be a short delay after your account creation before you can book equipment with this system. Once your account is functional, you will be able to book instruments and review reservations with a much more sophisticated interface.
