Metabolic flux and enrichment studies using MIMOSA
In this subcore, parameters of cellular and tissue metabolic flux are assessed by incorporation of stable isotope-labeled tracers (e.g., carbon-13, nitrogen-15, or deuterium) into metabolism. The CMC uses the targeted mass isotopomer multi-ordinate spectral analysis (MIMOSA) platform to interpret stable isotope labeling patterns for calculation of rates of discrete steps in glycolytic and mitochondrial metabolism. Such measurements can be used to identify differences in fuel usage (e.g., glycolysis vs. beta-oxidation), and non-oxidative (reductive carboxylation, anaplerosis and cataplerosis) contributions to the TCA cycle.

Islet isolation and function
In this subcore, our technical expertise in isolating rodent islets including rats, mice, and most recently ground squirrel from the pancreas ex vivo allows researchers to study the intact islets for their studies or to proceed with substrate-stimulated insulin secretion functional studies using our BioRep Technologies Islet Perfusion Systems (8-channel and 12-channel). For non-rodent islet studies, human donor islet procurement assistance as well as functional studies are also offered.

Cellular and mitochondrial respiration and bioenergetics
Cellular and mitochondrial respiration (OCR) and acidification (ECAR) are assessed with the Agilent Seahorse XF Pro 96. In a 96-well plate format, real-time measurements of cellular respiration, oxidative phosphorylation, mitochondrial leak, and mitochondrial transport are assessed as functional kinetic measurements of cellular bioenergetics. Changes in acidification are also observed simultaneously. An integrated Agilent BioTek Cytation5 imager correlates cell number via Hoechst nuclear stain to respiration for more accurate normalization of respiration.
