About the core

The Yale Lipidomics Innovation Core (YLIC) offers lipidomics analytical services, including sample preparation, lipidomics data acquisition, data analysis, and data consulting. We use an untargeted LC-MS/MS approach and identify most major lipid species. 

On request, we measure the absolute quantity of select lipid species in samples, including LPE, PE, ether PE, PG, PI, PS, cholesterol, cholesteryl ester, ceramide, glucosyl/galactosylceramide, lactosyl(hex2)ceramide, sphingomyelin, LPC, PC, ether PC, DG, ether DG, TAG, and ether TG. 

We are actively incorporating lipidomics research and method development with Dr. Kallol Gupta of the Department of Cell Biology. We also work closely with other cores, including West Campus Analytical Core and Chemical Metabolism Core. Feel free to contact us to find the core that best suits your research.

Services

Sample preparation

  • The core staff extracts lipids from user-provided samples. 
  • Cell pellets, in vitro assay materials, or tissue can be processed. If you are interested in using other types of samples, please contact us.
  • The user can provide extracted lipids. For consistency and equipment maintenance, please contact us to confirm detailed information about your extraction protocols, solvents, and materials. 

Data acquisition

  • The core staff uses LC-MS/MS to acquire chromatograms and spectra of the lipidome. Experiment quality and sample quality will be assessed. 
  • If the sample quality issues are found, the user will be asked to provide another sample. 
  • If quantification of minor lipids is requested, multiple sample preparations and data acquisitions will be ordered. 

Data analysis

  • Lipids are identified and/or quantified (as requested) with high confidence using public, in-house, and/or commercial MS libraries. 

Data consultation

  • Users may request consultation to discuss data analysis and learn sample preparation and lipid analysis methodology.

  • Orbitrap Exploris 240 mass spectrometer
  • Q Exactive UHMR Hybrid Quadrupole-Orbitrap mass spectrometer
  • Vanquish Neo UHPLC system
  • EASY-nLC 1200 system

West Campus Analytical Core is closely integrated with our core and expands our lipidomics-optimized instrumentation, including:

  • Agilent 1290 InfinityII LC - Agilent 6546 qTOF
  • Agilent 1290 UHPLC - Agilent 6550A qTOF

Chemical Metabolism Core provides services and facilities for metabolomics and targeted analysis.
