The Yale Flow Cytometry Shared Resource provides comprehensive flow cytometric analysis and cell sorting services to the Yale School of Medicine (YSM) community. The facility provides access to a broad range of well-maintained analytical and cell-sorting instruments, with dedicated staff support for cell sorter operation. In addition to instrument access, the facility provides user training, assay and technique development, protocol guidance, and several wet-bench and fee-for-service offerings, including Luminex-based assays, sample cryopreservation, staining, and data acquisition.

The Yale School of Medicine campus houses multiple models of analytical flow cytometers, including two CytoFLEX LX instruments (5 lasers, 20 fluorescence detectors), one BD LSR IIanalyzer (4 lasers, 18 fluorescence detectors), three BD FACSymphony cytometers (one with 5 lasers and two with 6 lasers, including a 780 nm laser; 28 fluorescence detectors), two BD Fortessa analyzers (5 lasers, 28 fluorescence detectors), and two Cytek Aurora spectral analyzers (5 lasers). The facility also maintains an Amnis ImageStream Mark II imaging flow cytometer and a Luminex IntelliFLEX DRSE analyzer.

For cell sorting, the facility operates a Sony SH800, two BD FACSAria sorters with full-time operator support, and a BD FACSAria Fusion. A Sony MA900 cell sorter is independently operated within the BSL-3 facility, enabling the safe sorting of samples that may contain potentially infectious agents. The facility also has two ThermoFisher Bigfoot cell sorters and a Cytek Aurora cell sorter, providing the flexibility to sort using either spectral unmixing or traditional fluorescence detection with compensation. Despite their high-speed sorting capabilities, the Bigfoot sorters are exceptionally gentle on cells and offer the added advantage of simultaneous sorting into any combination of 1.5 mL, 5 mL, 15 mL, or 50 mL tubes.
