Microbiology Service
About the service
The microbiology service performs aerobic and anaerobic bacterial culture, identification, optional antibiotic sensitivity and resistance, and fungal isolation from blood and tissue specimens from animals within the Yale vivarium. Examination of fur and fecal samples for ectoparasites and endoparasites, respectively, is available and can be requested.

Services
Cellular and mitochondrial respiration and bioenergetics
Cellular and mitochondrial respiration (OCR) and acidification (ECAR) are assessed with the Agilent Seahorse XF Pro 96. In a 96-well plate format, real-time measurements of cellular respiration, oxidative phosphorylation, mitochondrial leak, and mitochondrial transport are assessed as functional kinetic measurements of cellular bioenergetics. Changes in acidification are also observed simultaneously. An integrated Agilent BioTek Cytation5 imager correlates cell number via Hoechst nuclear stain to respiration for more accurate normalization of respiration.

Analyze cells from tubes and plates with 4 lasers (violet, blue, yellow/green, red) and 14 detectors (6 violet, 2 blue, 3 yellow, 3 red).

BD Symphony S6 Cell Sorter
Sort into 5 mL round tubes, 15 mL conical tubes, or plates (24-, 48-, and 96-well) with 5 lasers (UV, violet, blue, yellow/green, red) and 23 detectors (6 UV, 6 violet, 4 blue, 4 yellow/green, 3 red).
